I think you can enjoy the mystery as a standalone. But it's a much richer experience if you start at the beginning and work you way through. One of the best things about this series is the development of the characters and the growth and expansion of the In Death world.
I started with a book that was maybe the 15th in the series, and I enjoyed it. I think I read another one or two out of order. But I got a lot more enjoyment out of each book once I went back to the beginning and started there.
If you're just starting out, I envy you. Wish I had the opportunity to read them all again for the first time! (I've reach each one at least three times.)

You absolutely CANNOT read this series as a stand alone. In order to understand the characters and their stories (especially Eve and Rourke), you have to start with book one. All of the characters are connected somehow and if you skip through the books, you will be lost and confused when you move to another book. Just start with book one. You will not be disappointed.

I picked up again about book #45 or so and didn't feel like I missed much of the storyline. A few things are referred to that appeared in previous books, but they are mentioned in a way that I could easily figure out what I had missed. I have stayed up with the releases since about book 45 and have been enjoying the series even with the big gap.
They are fun to read from the beginning, though.

I think it might be better to start from the beginning to show why character do the things they do. My opinion.

It is better if you have, you can read the book as a mystery by itself but to really get into the main character/s you should read the other novels. All of the main character have their own growth and histories throughout the series and you learn more about them as you read each book.
So it's really up to you whether you want to get a lot out of the series. If you wanted you could begin with the first one and see how you like it, or wait for this one, read it and see how it feels to you and go back and read the rest.
