Under the influence of sub-bacteriostatic doses of novoimanin on agar two types of colonies of staphylococcus arose – normal size and forms, consisting of the usual cell size, and in a larger number – dwarf colonies consisting of larger cells – round, oval, rod-shaped, solitary, pairwise or small groups unevenly colored; Streptococcus also lost their inherent location, they are larger, their tinctorial properties are changed.


In Bacillus subtilis, changes in the shape of colonies are more noticeable, and in Lactobacterium Plantarum (not in all strains) – cell forms. All these phenomena are less pronounced than, for example, in the same bacteria under the influence of penicillin, but still very vivid. Pseudomonas aeruginosa in the presence of novoimanin in the medium grew in the form of small bespigamentnyh colonies. It is interesting to note the coincidence of these data with the observations of PS Volosovets (1964), who noted the loss of pigment by the Pseudomonas aeruginosa, isolated from patients treated with a new Lyman.


Morphological changes in bacteria under the influence of the new immanin were also found in other species of bacteria, especially in the presence of serum in the medium. The change in the morphology of cells under the influence of the new immanin depends on the nature of the medium and the amount of the preparation in it, the time and conditions of contact. But even when the influence of these factors is taken into account, a certain regularity is revealed: the newiman causes an increase or decrease in cell sizes, a change in their tinctorial properties; in the study of the majority (only Gram-positive species), a decrease in the chromatin substance.


Novoimanin, according to GI Poroshina (1964), causes inhibition of acid formation in lactic acid bacteria. The active acidity of the medium with their growth in the presence of subbacteriostatic doses of the antibiotic in the medium increases more slowly than without it. The fermenting properties of Leuconostoc mesenterioides, Streptococcus lactis, Lactobact also change . plantarum; on a medium with an antibiotic, more acetic acid is observed than in a medium without a preparation, i.e., a new immanin causes a shift in the fermentation process of lactic acid bacteria towards heterofermentation. Novoimanin inhibits the rate of dehydrogenation of glucose, galactose, fructose, sucrose, arabinose, xylose, mannitol, sorbitol, alcohol, α- and β-alanine, leucine in L eucopo sto with те s е nter ioides, and inStreptococcus lactis only glucose, lactose, sucrose, maltose, sorbitol, in Lactobact. r1ap1a rum α-alanine. Differences in the character and degree of influence of the new Lyman on the dehydrogenation of substrates of not only different species but also strains of lactic acid bacteria are noted.

Influence of new lyman on tissue regeneration

Reduction of microflora in pus wounds in the treatment of patients with novoimanin PS Volosovets (1964b) noted already in the first days of antibiotic application – in 2 – 3 days the number of colonies sown from pus was significantly less than before the beginning of treatment; after 5 days, microbes in the crops were not detected in 51.6% of patients, in 10 days in 83.7% of patients; from pus disappeared, not only gram-positive, sensitive to novoimanin, but also some gram-negative bacteria (E. coli).


 





In the treatment of neuromanni patients, from pus wounds of which the associated flora was sown (Staphylococcus aureus, hemolytic streptococcus, Escherichia coli), staphylococcus disappeared later than other species. But Proteus vulgaris, and Ps. aeruginosa the effect of the antibiotic was less pronounced; Combining it with colimycin or polymyxin M led to favorable results in all cases. In the course of treatment with neovymanin, pathogens sown from the foci showed changes in the size and shape of the colonies of staphylococci, the latter losing their bunching habit and the change in tinctorial properties. Morphological changes in the cells during the treatment with neomycin were also observed in the Pseudomonas aeruginosa and vulgar protein; the last on the 10th – 15th day of the application of the new lyman lost creeping growth, and in Ps. aeruginosa weakened the ability to pigment.

An investigation into the dynamics of the influence of the new Lyman on the regeneration of tissues showed that along with a decrease in the number of microbes, the growth of fresh granulations and epithelization increased.Cytological studies have shown that already on the 3rd – 5th day after the beginning of the application of the new manganin, macrophages appeared, in a large number of playblasts; phagocytosis increased; the number of regenerative cells gradually increased, degenerating – decreased. The intensification of regenerative processes was observed in all cases and corresponded to the dynamics of microflora, the clinical picture of the pathological process, which usually resulted in complete recovery by the 10th day, sometimes even earlier.

AF Burianov (1968) describes the use of neimannin in the complex treatment of neonatal mastitis (in 46 children). The author notes that in recent times the severe forms of mastitis caused by ayatibiothoracoresistant staphylococci, which are of the type of phlegmon of newborns, have become frequent. They are characterized by a sharp onset and rapid spread of the process, and necrosis of the skin and subcutaneous tissue may occur. Novoimanin was used by electrophoresis; after surgical intervention, the cavity was drained with tampons moistened in a new immanin solution (1% alcohol solution of neimannin was diluted 1:10 with 10% glucose solution). To prevent complications of the upper respiratory tract, novoymanin was used as an aerosol. After application of complex treatment on the second-third day, the temperature normalized, the general condition improved child.

At the same time, the wound was cleaned, granulations intensified, and the size of the wound decreased.Bacteriological study confirmed the clinical effect. The author considers the use of newiman in the complex treatment of neonatal mastitis an effective means of prevention and treatment. MN Kovalishin (1967) used the newimanin in 86 patients in the orthopedic and traumatological department. The drug was put under gypsum bandages to wounds in the form of lotions for a long time. At the same time, a good tissue regeneration was observed; skin irritation or other complications not noted. In the author’s opinion, the newimanin is a good auxiliary agent of local action in the complex treatment of patients.

Detection of medicinal properties of imanin

The identification of the ability of imanin to inhibit the multiplication of plant and animal viruses has raised the task of determining which components of the drug determine its antiviral properties. In this connection, flavonoids and tannins of imanine were obtained and investigated, their effect on plant and animal viruses was studied (Derbentseva et al., 19726).


The basis of the scheme adopted for the preparation of these substances is the principle of exhaustive extraction of the material under study by various solvents – ether, ethyl acetate, acetone and water, aimed at isolating groups of substances with different degrees of polymerization.Thus, ether extracts contain monomolecular polyphenolic compounds, ethyl acetate extracts – easily hydrolysed tannins, acetone extracts – hardly hydrolyzed tannins, hot water extracts – water-soluble tannins and free carbohydrates, and finally a water-insoluble residue, which is highly condensed substances – flobophrenes.


Identification of the substances that make up each fraction was carried out by chromatographic analysis on paper, qualitative reactions to flavonoids and tannins, and removal of absorption spectra in the UV region. The hydrolysable tannins present in the ethyl acetate and acetone fractions underwent acid hydrolysis, resulting in the production of polyoxyflavone aglycones: quercetin, 7-hydroxyflavone and 2,4-dioxiflavone, as well as free fructose, glucose, galactose and rhamnose sugars. Catechins are found in the water-soluble portion of the ethyl acetate fraction and in hot water extraction. When comparing the content of tannins in the herb of St. John’s wort and imanin, it was found that the composition of all fractions extracted by organic solvents, aqueous alkali and water is not the same. For example,


The study of the antiviral activity of the obtained imanin fractions was carried out on the influenza virus in vitro by a contact method. The fractions that showed the greatest activity in these experiments were tested in in ovo experiments on chick embryos. In contact experiments, the influenza virus type A strain PR-8 was used; in experiments in ovo, influenza virus type A2 strain 2226. Virulicidal activity is most pronounced in the ether-soluble fraction comprising monomeric polyphenols, in the acetone fraction and in the water-soluble portion of the ethyl acetate fraction containing catechins and flavonoid agglucones, and in the fraction recovered by hot water, which also has catechins are found.These substances in vitro inactivate the virus group by 95-100% to the same extent as the imanin itself. The activity of the remaining fractions is less and their inactivation of the virus does not exceed 50-70%. In experiments in ovo on chicken embryos, the same fractions containing catechins and flavonoid aglycons are most active, which, like Imanin itself, completely inhibit the accumulation of viral agglutinins. The ability to inactivate the influenza virus in similar fractions obtained from the herb of St. John’s wort is somewhat weaker (50-70%).
