I start with a character sheet I created years ago with the basics: physical description, special skills and talents, fears and weaknesses, important prior events/experiences, immediate family & other important people. Then as I write, the characters start revealing more and more about themselves. By the end of a book I know them very well, at which point I go back and revise, part of which is making sure each character is consistent with whoever they've evolved into. :)
