Ew99 You should absolutely not read it first. There are major plot points that depend on familiarity with the characters, with previous occurrences and with previous storylines.
Rowena Hoseason I wouldn't start here. It's brilliant, but so much of what is enjoyable about 'Overture' are the references to other events in the story cycle. I think that if you've not read any of the others then this will be confusing and overwhelming. Better to start read the original series then come back to Overture, which fills in many of the gaps...
Martin I've never read Sandman and started with Overture - and I would not recommend to do so. It was very clear that there are many references that I didn't get. Cannot comment for the rest, but I was not impressed by this story.
Anjan I would like to think the series - like dreams - exists beyond the need for chronology. There are no spoilers.

Cantatis If you read The Sandman Overture, I guarantee you'll want to read the whole series; and since you're going to want to read the whole series anyway, I recommend doing that first because it's the order Gaiman intended. Besides, the Overture explains a lot of what happens in the series, but the reverse isn't always true.
Daniel While it could be read first, as it does take place chronologically before the events of the wider story, it probably is best to read the original series first. There are a lot of Easter eggs and cameos, and even semi-spoilers for some of the other stories.

shean I just finished this and I've been wondering the same thing myself, whether it'd have the same impact if a new Sandman reader were to pick this up before taking the journey through the reset of the series first... (because I only have my experience of the magic of it all, and this came last)

I think the answer is yes, that this stands on it's own as an overture. The beginning of what follows. I'd encourage anyone, at any point in their Sandman journey to make their way through this one.
