I haven't read Sweet Thursday or Cannery Row. Should I read them in order (Cannery Row and then Sweet Thursday) or does it matter?

The "purists" who suggest Cannery Row first should also add The Sea of Cortez, and The Logbook from the Sea Cortez to get a full picture of the "Doc". I had the privilege of actually getting to visit his lab in Monterey. Doc and Steinbeck's spirits were everywhere, as were the spirits of other great writers, cartoonists, artists, and musicians who used his lab later as a "Bohemian Salon and Social Club".
