Along with expertise in specific scientific fields, do you think a good sense of humor (like that of Mark Watney's) is a quality that should be actively sought after in potential real-life Martian (or other new-frontier) explorers?

Andy Weir Yes, definitely. A good personality is critical. Not for the scientific aspects of a mission, but in order to get along with crewmates. A manned mission to Mars requires the crew to spend literally years together in cramped quarters.It is critical that they get along.
