Most of this advice is... not helpful, but I really like this quote: “All characters (just like people) have this hole right through their middle and all of them are desperately trying to hide it and live with it.” Really succinct way of putting something that feels obvious, in a way that’s less straightforward than assigning characters a “fatal flaw” or a “driving want.”
