First breakthrough in gene therapy against aging
Published on April 25, 2016

In September 2015, then 44 year-old CEO of BioViva USA Inc. Elizabeth Parrish received two of her own company's experimental gene therapies: one to protect against loss of muscle mass with age, another to battle stem cell depletion responsible for diverse age-related diseases and infirmities.

The treatment was originally intended to demonstrate the safety of the latest generation of the therapies. But if early data is accurate, it is already the world's first successful example of telomere lengthening via gene therapy in a human individual. Gene therapy has been used to lengthen telomeres before in cultured cells and in mice, but never in a human patient.

Telomeres are short segments of DNA which cap the ends of every chromosome, acting as 'buffers' against wear and tear. They shorten with every cell division, eventually getting too short to protect the chromosome, causing the cell to malfunction and the body to age.

In September 2015, telomere data taken from Parrish's white blood cells by SpectraCell's specialised clinical testing laboratory in Houston, Texas, immediately before therapies were administered, revealed that Parrish's telomeres were unusually short for her age, leaving her vulnerable to age-associated diseases earlier in life.

n March 2016, the same tests were taken again by SpectraCell revealed that her telomeres had lengthened by approximately 20 years, from 6.71kb to 7.33kb. This implies that Parrish's white blood cells (leukocytes) have become biologically younger. These findings were independently verified by the Brussels-based non-profit HEALES (HEalthy Life Extension Company), and the Biogerontology Research Foundation, a UK-based charity committed to combating age-related diseases.

Parrish's reaction: "Current therapeutics offer only marginal benefits for people suffering from diseases of aging. Additionally, lifestyle modification has limited impact for treating these diseases. Advances in biotechnology is the best solution, and if these results are anywhere near accurate, we've made history", Parrish said.

Bioviva will continue to monitor Parrish's blood for months and years to come. Meanwhile, BioViva will be testing new gene therapies and combination gene therapies to restore age related damage. It remains to be seen whether the success in leukocytes can expanded to other tissues and organs, and repeated in future patients. For now all the answers lie in the cells of Elizabeth Parrish, 'patient zero' of restorative gene therapy.

Since her first gene therapy injections BioViva has received global interest from both the scientific and investment communities. Earlier this month BioViva became a portfolio company of Deep Knowledge Life Sciences (DKLS), a London-based investment fund which aims to accelerate the development of biotechnologies for healthy longevity.

Dmitry Kaminskiy, founding partner of DKLS, said "BioViva has the potential to create breakthroughs in human gene therapy research, while leapfrogging companies in the biotech market."

J.D. Lovil (jd_lovil) | 85 comments Since aging is essentially a 'clearing the field' add on process for species, I suspect that modern science will find it relatively easy to stop the aging process, with nanotech, neuro and bio chemistry and gene mods. The real question is, what are we to do with the rising population of immortals? Even exporting them to space colonies will hit a singularity, where they cannot be exported as fast as the population grows.

Elisabet Norris | 486 comments The article doesn't really explain how they are lengthening the telomeres.
It's actually a gene therapy with the use of Adeno Associated Virus that makes a copy of itself and attaches itself onto a specific location on chromosome 19 where it makes copies of its DNA.
The variant of this virus that they use has been genetically modified and includes a payload of therapeutic DNA ....this way, the virus can only enter a cell, but not reproduce or enter any other cells.... this is good because it won't be able to cause disease.

So, I have a couple of questions:
1. Since the virus can only "infect" one cell, how do we determine or how are we able to select which type of cell it should enter? How do we control it?
2. What if more than one virus tries to enter the same cell? Does that mess with the coding? Does it mess with the dosage?

James Morcan | 11378 comments Further to Lisa's questions, my main question would be relating to the process of lengthening telomeres to defeat or slow aging...I have previously read all humans and animals who live a long time have been shown to have longer telomeres.

So basic logic would assume lengthening telomeres via gene therapy or any other method would equal either physical immortality or at least a much longer life of hundreds or thousands of years. However, I have read that it could actually shorten one's life as early scientific research has indicated forcibly manipulating telomeres in humans would drastically increase the chance of developing cancer.

So I'd like to know how the scientists involved in this new gene therapy model are proposing to get around that.

Here's perhaps a more objective article on this development:

Scientists claim they've completed the first successful gene therapy against human ageing -- http://www.sciencealert.com/scientist...

Some excerpts include:

"Originally meant to prove that her company’s gene therapy was safe, the results - should they prove to be effective in the long-term and withstand due scientific scrutiny - would be the very first successful demonstration of telomere lengthening in any human."

"Telomeres are short segments of DNA that are found on the ends of each chromosome. These act as 'buffers' for the wear and tear of natural ageing. But with sustained cell division, telomeres eventually get too short to protect the chromosome. When this happens, it causes the call to malfunction and leads to ageing."

"The basis for the success of Parrish’s gene therapy is related to the telomere scores - which are calculated based on the telomere length in white blood cells (T-lymphocytes). Higher telomere scores indicate 'younger cells'. Compared to average T-lymphocytes of the American population within the same age range, 44 year old Parrish claims that the gene therapies she underwent worked and showed that it reversed 20 years of telomere shortening."

As far as I understand, it has been considered increasing the chance of cancer in the earlier gene therapies. This is because of the location/site of insertions of the virus. It actually disrupted our DNA.
But, the type of gene therapy they are doing: Adeno Associated Virus (AAV)...their target sites are safer and don't cause any harm to our DNA.

I'm open to that belief that aging is more mental conditioning. For example, I heard blind people age slower because (according to some theories) they aren't looking in the mirror all the time thinking things like "gee, I look good for 40" or "I'm 39, I wonder when I'll go grey" etc.

However, to play the Devil's Advocate, it seems everything on this planet ages and dies.
So, with this idea that aging is just a mental construct or due to a belief system, how do we explain animals who also age and then die?

Elisabet Norris | 486 comments What's interesting is that in mice, the telomeres don't seem to have the same effect on them as it does on humans.... as their telomeres shorten, or even disappear, the mice still live on.

A study was done in 1997 (so could be old news....) that studied the involvement of telomerase in in vitro immortalization of human cells. It says that this in vitro immortalization of tumor viruses occurs in two phases:
1. Extension of its lifespan. The culture goes into a crisis period.
2. Escaping crisis of a rare cell in the culture, which goes on to proliferate indefinitely.

Their hypothesis was that the actual shortening of the telomeres acted as a signal of age deterioration and crisis. Therefore, the cell needs to activate telomerase to survive these stages.

Often, they found that the telomeres kept shortening during life extension period and there was no telomerase detectable. The cells that survived the crisis, had activated telomerase and had a stable or lengthened telomeres.

We have various cell lines, so this doesn't apply to all of them. In the study, about 1/4 of our in vitro immortalized cell lines, had no detectable telomerase activity, but they still have very long telomeres. These are called Alternative Lengthening of Telomeres (ALT).

The fact that the cells don't need telomerase to maintain the telomeres, means that the cell line is using something else.

Since immortalization rates differ between races, it seems to me it would be hard to find the "cure" for cell death by using murine cell lines.

This makes me wonder... if there's a way around telomeres..... could we possibly find a way to renew our cells and voiding/bypassing the telomere's senescence all together?

PARABIOSIS AND THE “ELITE” https://gizadeathstar.com/2017/01/par...

Written by Joseph P. Farrell

This very important article was shared by Ms. S.H., and it's one of those articles that compels attention, and sharing here.

But first a little background. My co-author Dr. Scott deHart and I noted in our book Transhumanism: A Grimoire of Alchemical Agendas, that various schemes were being pursued by the super-wealthy and their subsidiary technocratic "elites" to transform mankind into... something else. Chief among these pursuits and schemes are various ideas to render humans immortal, or to come as close as possible, via scientific and technological means. The schemes range from downloading and uploading a person's memories and so on, into a computer, to cloning, genetic and nano-medicine designed to combat diseases and repaid body tissue and organs, to 3-D printing and manufacture of organs, and so on. Some transhumanists speak openly of the prospects of virtual immortality by these means, endless clones of an individual whose personality "emgrams" are downloaded and uploaded.

And among the means being pursued for this "alchemy of immortality" is parabiosis:

Parabiosis: Elite Transfusing Blood of the Young? Original Research Funded by Rockefeller Foundation

The essence of the concept, as reported in the article, first was reported in an article in the scientific journal Nature, which stated the following:

In the past few years, however, a small number of labs have revived parabiosis, especially in the field of ageing research. By joining the circulatory system of an old mouse to that of a young mouse, scientists have produced some remarkable results. In the heart, brain, muscles and almost every other tissue examined, the blood of young mice seems to bring new life to ageing organs, making old mice stronger, smarter and healthier. It even makes their fur shinier. Now these labs have begun to identify the components of young blood that are responsible for these changes. And last September, a clinical trial in California became the first to start testing the benefits of young blood in older people with Alzheimer’s disease. (Emphasis added)

And there you have it: the "blood of the young" apparently rejuvenates older tissues when injected into a compatible organism, in this case, mice. The article raises some disturbing questions, but there is a statement that caught my eye, and I suspect it caught yours as well:

McCay’s experiments were originally funded by the Rockefeller Foundation. Scroll down the sample page and note the study was done with grants from the Office of Naval Research in conjunction with none other than the Rockefeller Foundation. It seems this organization has it’s fingers in every little pie. (Emphases in the original)

Rockefeller, and ONR(The Office of Naval Research). While nothing surprises me any more about the ghoulish and macabre activities funded by the Rockefailure foundation - after all, these geniuses were the ones who also gave us agribusiness, and with that, ultimately the GMO, and let's not forget their involvement in the 1930s eugenics research, progressivist "edgykayshun" and a whole host of other nutty ideas - what caught my eye here was the Office of Naval Research. While doing our research for our book on Transhumanism, Dr. deHart uncovered something that to this day perplexes me, and that was the discovery that the Department of Energy at the Oakridge National Laboratories in Tennessee was sponsoring research into the genetic basis of a list - and a very odd list it is, too! - of traits, from genetic predispositions to alcoholism, perfect musical pitch, tongue curling, ear wiggling, and homosexuality! One wonders indeed what any of these things have to do with energy, and why one would be researching them at a nuclear energy laboratory.

What unites these bizarre stories is the connection to the military, and to the "elite." All of that suggests that these things are being researched for one simple reason: they are perceived as avenues to the expansion of their power, and certainly longevity and control of its means would be one such expansion.

But another thought occurred to me as I read this article, and herewith my high octane speculation of the day, for I cannot help come away from this article feeling - perhaps a better word here would be intuiting - that there is some sort of deep connection between the phenomenon of parabiosis, and all the stories we've seen in the past few decades, of pedophilia scandals and child sex-slavery rings, from all over the world, stories that have ranged the whole spectrum, up to and including occult rituals and human sacrifice. Certain members of the "elite" do seem oddly long-lived, even as their faces sag like melting wax on a wax museum replica. And some of these have publicly expressed an interest in parabiosis.

Of course, it's purely speculation. I know of no detailed evidence of any sort to connect the two stories. But then again, there have been people who have suspected such connections. And they end up vilified, persecuted in various ways, laughed at, or simply dead by all-too-convenient accident. And perhaps that information about ONR and the Rockefailure foundation is a clue as to why. The article ends on this note, and it echoes my own suspicions:

This is all very disturbing and not pleasant to consider by any means. The logical deduction does not lead to a good place. An industrial procurement system would have to be established cataloging the type of blood, it’s origin and vintage(age). We know that ethics are no boundary to the elite throughout history. Any rational person would have to ask themselves this question. How long before the appetites of the rich and powerful create a market for the blood of the young? If they haven’t already.

Feliks (dzerzhinsky) Absolutely not; and there is really no reason why any sane person would (or should) want such a thing. Fear of aging is absurd and silly. Perhaps it stems from the current fad for sexy 'vampires live forever'; nevertheless it is still daft.

Are people truly so empty inside; so devoid of inner depth, lacking in loving relationships, and achieving of no personal accomplishments.... that they throw everything into preserving their plastic mannequin looks? As if it is all they have left to value in themselves? This is a society addicted to image rather than substance.

Only morons want to stay physically young past a normal human timespan. Its strictly for sick, dizzy bimbos like Joan Collins and Marlene Dietrich. Self-obsessed movie stars and other entertainment-industry drips. Like Norma Desmond, in 'Sunset Blvd'.

Well, too bad for all such dopes & simpletons but eventually you have to undergo your Moment of Truth, recognize your Mortality, and Meet your Maker. No one can do it for you, you got to walk that valley by yourself. We're all made from the same clay.

No one stays James Dean forever. Liz Taylor eventually gets fat and Mont Clift loses his face sooner or later. Normal wear and tear! Get over it!

Fortunately the laws of physics will never allow any other expedient. It is impossible for anything to 'not age'; thank God!

 B. Sinsational (bsinsationalx1f339) | 40 comments If i may be blunt, let me point out a few facts.
If genuine immortality is achieved it will never be told or avaliable for the general population.
The Elite with all the money, facilities, and scientist at their beck and call are the ones that would be keeping that within their circles.
But one should also be aware that they seem to adhere to their own version obligated fair notice, what form they choose tto tell this by is totally along with their devious nature. They (geneticallly prooven) to come from the wizards and fortune tellers that resided in the temple, but the magic is words, and images, they hold power, (consider an illusionist ..) they put it with a twist in front of peoples noses and no one gets that thats what they do..
On immortality:
Immortal cells was discovered many decades ago they are called HeLa cells.
Gmo babies have been manufactured for a long time, they were about 25 years when they went public, partially because it wasnt approved. That was 2012. (Where does the soul come from?)
So their science is about 25-30years ahead of what they have been telling us. I know (due to a person involved in the project) that already in 1998 they had androids that they let out on various assignements on the streets, etc. They had at that point already done several years research on transfering conciousness but had only partial success.
But in 2002 their research got a slightly different direction and security was extreme.
What I have found was that they did a discovery in egypt some time early 90's and that it put things in a different light. The ancient egyptian gods had intimate interactions with humans, but they feared loosing part of their conciousness (soul?) so they used very special "devices" that they transfered their conciousness/soul to for safe keeping. (Kolbrin the original binle speaks very detailed about this as does a lot of the sacred texts that have been found), more specifically they used a device they ALL called ankh of osiris.
When the gods wanted to regenerate they stored the soul/conciousness in the device as well. The texts warn that the ramifications for a human to use that, or let that soul posses them will be dire, rapid aging, memory disturbances, and even dementia would be the punishment.
Side note: This artefact is and has been subsequently used in the second ritual of initiation (that is not for public eyes) that they hold for the newly elected president.
Part of this knowledge is not anything you would find on the net, it is among others what is told by archeological experts who found the grave of osiris, linguistic experts who worked on the translation, neurologist who have been working on this, etc (and high inside sources).
The Ankh was said to have the ability to move a conciousness from one body to another, but caution was urged since the conciusness that originally belonged there might object, interfere, etc and cause severe meental illness. Tests were done using it in combintion with what is known as devils breath.
Remarkably, on a very very obscure movie (Osiris films) company that just did that one film about a man who is hunted by memory flashes, and finds out that his conciousness used to be in a different body, and the new body was grown in a lab. Expensive fx and soecial effects, obscure actors no one ever heard of, no publicity, it just faded away.. and notthing of it is even searcheable. (Was sent to me).
Transferring of your conciousness would qualify as immortality id say.
As for the rest of humanity, healthy people dont generate money past a certain age, so artificial substances are added to guarantee we get sick and die in a timely fashion.

Another sidenote: it has thru science been determined that the conciousness/soul is a form of energy, it doesnt vanish naturally it just changes form, it was one of the biggest scientific discoveries they made at the same time as they found out that the 90% of the brain they thought was inactive actually was highly active, they were just using the wrong means to measure it.
What it uses is scalar waves, and scalar energy is one of the strongest, fastest most powerful things in the universe. It is literally what shapes our reality, it is what constructs a reality that we with our fysical manifestations very limited abilities can comprahend.
(Height, widths, Distances, time, movement etc) that is part of the knowledge the ancient mystcs have always known.
Additional info that gives a lot of knowledge in this area can be found if you research Tesla and the scalar heart.

Ian Miller | 1422 comments HeLa cells are cervical cancer cells taken from Henrietta Lacks, hence the name. Henrietta died, but the cancer cells are particularly durable, and they have been used in scientific research because they are a sort of standard, and they are relatively old, as Henrietta died in 1951. They ar not fungi - that are cervical cancer cells.

Okay, so HeLa cells are immortal cells but cancerous?
Can't you scientists somehow remove the cancerous part of those cells, Ian? :)

James Morcan | 11378 comments Really Jeffrey?
So when does our immortality option become available in regards to our lives?

Those researchers would have a better answer.

Remarkable since many scientist actually says cancer is fungus. Even Otto Warburg who got a nobel price for tgat. Eric enby is another there are numeral.

Ian Miller | 1422 comments Cancer is most definitely the uncontrolled reproduction of cells that carry much of the host's DNA but has mutated in some way. Every cancer is therefore different from any other in fine detail, although of course it carries part of the information of the organ where it started, thus breast cancer contains some breast cell information, it contains information specific to the host, and because certain mutations are more common than others, there are various strands of cancer. Thus some breast cancers are held back by oestrogen inhibitors. Cancer is most definitely NOT a fungus.

Might that be true for people, too?

Ian Miller | 1422 comments Lance, I think that is only temporary. I think that was a trick used illegally by some sports people when steroids, etc are too risky.
