According to her twitter she plans on doing a Kickstarter to publish Spindle sometime this year (2017).

I just did a search for "Prestor John" and "Spindle of Necessity" on Valente's blog, and the last mention of it was in 2012 when she was talking about needing to find a new publisher.
She says in that post that she's dedicated to publishing it someday - but it has been 4 years and no mention of it since then. The odds seem low at this point, but you could try asking her via her blog/website (she doesn't seem very active here).
