Is it necessary to read the prologue before starting the actual book? The prologue gives a brief history of the hobbits. I have not read The Hobbit, therefore will I be able to understand if I directly start from Chapter 1?

ATrueBookworm The prologue is a history of the hobbits and how the ring came to them. If you have not read The Hobbit then it is a good idea to read the prologue. Things happen in the book that you will not understand unless you know the history of the Ring and and how a hobbit who came to possess it. If you do not care or are planning to read The Hobbit after The Fellowship of the Ring, than it would be safe to skip the prologue. Otherwise, I would advise reading it.
