The main character is a female inspector trying to stop the killing or replacement of characters in books. I believe the premise also included if the book stopped being read the book and the characters within it were dissolved into individual letters and nobody could read the book again. When a book was being read the characters would behave much like actors on stage when their passage was being read.
Read it around 2011.

I second Kat's opinion... This does sound a lot like The Well of Lost Plots, the third book in the Thursday Next series.
