Which should I read first, Murder on the Orient Express or Murder in Mesopotamia? A couple lists I have found said Mesopotamia, but the list on the Agatha Christie website recommended Orient Express. Which one should I read first?

Either way would be fine. The two books are not connected to each other. In fact, most of Agatha Christie's books are standalone, and can be read in any order. Just pick up whichever one looks more interesting to you. They're both very enjoyable books (though Orient Express is more famous and more intriguing).
I've read them pretty much out of order, I think, except for the Tuppence and Tommy books. As Mitali said, in this case it won't matter, they're completely separate. Had I not started reading her as a kid, just grabbing up whatever books I could, I probably would have tried them in the order they were published, but they really pretty much stand-alones, over all.
