I think the key difference is that Harley is a protagonist on an equal footing with Pamela. She has her own goals and an independent life, and her appearances are teamups. Janet is in her own words a sidekick, and her relationship with Pamela highlights a lot of Pamela's flaws, especially her selfishness. It's a pretty toxic relationship and both of them know it, which is what makes it so interesting.
