ルーヴル美術館のソニエール館長が異様な死体で発見された。死体はグランド・ギャラリーに、ダ・ヴィンチの最も有名な素描〈ウィトルウィウス的人体図〉を模した形で横たわっていた。殺害当夜、館長と会う約束をしていたハーヴァード大学教授ラングドンは、警察より捜査協力を求められる。現場に駆けつけた館長の孫娘で暗号解読官であるソフィーは、一目で祖父が自分にしか分からない暗号を残していることに気付く……。
