The Color of Magic leaves off at a cliff hanger.

I would suggest reading The Colour of Magic first. I think this is considered as a two book story. This starts from where the other one left off. I will say that I think this book is better than the first.

You can make it through The Light Fantastic without reading The Color of Magic, but it does deal with the same characters and situation. Without meaning to spoil much, it will be very confusing for the first 20-50 pages, but then you can keep going without confusion at all.
You don't need The Color of Magic to get the plot of this story, but the main characters are not reintroduced, and The Light Fantastic does pick up where The Color of Magic left off.
On the other hand, the continuity and deep characterization probably are not the reasons you're reading the series.
So to make a very long post short: you can, you won't miss anything terribly important, but be prepared for a little confusion at the beginning because they fit together. The Light Fantastic is in no way a completely standalone book.
